Apple Proliferation Group

Group 16SrX phytoplasmas: Apple Proliferation Group (AP/PD/ESFY)

Phytoplasmas belonging to the Apple Proliferation group, or 16SrX taxonomic group (based on restriction fragment length polymorphism of 16S rRNA genes) comprises organisms associated with different diseases depending on the plant host.

"Ca. Phytoplasma mali" causes apple proliferation in pome fruit, "Ca. Phytoplasma pyri" is responsible of peer decline, while "Ca. Phytoplasma prunorum" is the agent of "European Stone FruitYellows" (ESFY).

Given their similarity, the molecular assay performed with IPADLAB kit enalbles detection of any one of those species. Normally a further analysis for distinguishing among them is not necessary, because every one of the three pathogens has host specificity.

Nevertheless, the DNA analytical fragment generated with the kit upon amplification of diseased samples can be subjected to RFLP analysis, as detailed in the instructions (reagents are not included). The RFLP pattern allows to distinguish between the analyzed species.
